In situ imaging of vortices in Bose-Einstein condensates

Laboratory observations of vortex dynamics in Bose-Einstein condensates (BECs) are essential for determination of many aspects of superfluid dynamics in these systems. We present a novel application of dark-field imaging that enables \texttt{\it in situ} detection of two-dimensional vortex distributions in single-component BECs, a step towards real-time measurements of complex two-dimensional vortex dynamics within a single BEC. By rotating a 87^{87}Rb BEC in a magnetic trap, we generate a triangular lattice of vortex cores in the BEC, with core diameters on the order of 400 nm and cores separated by approximately 9 ÎĽ\mum. We have experimentally confirmed that the positions of the vortex cores can be determined without the need for ballistic expansion of the BEC.Comment: 5 pages, 4 figure

Stress and Emotion Classification Using Jitter and Shimmer Features

    Get PDF
In this paper, we evaluate the use of appended jitter and shimmer speech features for the classification of human speaking styles and of animal vocalization arousal levels. Jitter and shimmer features are extracted from the fundamental frequency contour and added to baseline spectral features, specifically Mel-frequency cepstral coefficients (MFCCs) for human speech and Greenwood function cepstral coefficients (GFCCs) for animal vocalizations. Hidden Markov models (HMMs) with Gaussian mixture models (GMMs) state distributions are used for classification. The appended jitter and shimmer features result in an increase in classification accuracy for several illustrative datasets, including the SUSAS dataset for human speaking styles as well as vocalizations labeled by arousal level for African elephant and Rhesus monkey species

Insights Into The Recq Helicase Mechanism Revealed By The Structure Of The Helicase Domain Of Human Recql5

Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP)RecQ helicases are important maintainers of genome integrity with distinct roles in almost every cellular process requiring access to DNA. RECQL5 is one of five human RecQ proteins and is particularly versatile in this regard, forming protein complexes with a diverse set of cellular partners in order to coordinate its helicase activity to various processes including replication, recombination and DNA repair. In this study, we have determined crystal structures of the core helicase domain of RECQL5 both with and without the nucleotide ADP in two distinctly different ('Open' and 'Closed') conformations. Small angle X-ray scattering studies show that the 'Open' form of the protein predominates in solution and we discuss implications of this with regards to the RECQL5 mechanism and conformational changes. We have measured the ATPase, helicase and DNA binding properties of various RECQL5 constructs and variants and discuss the role of these regions and residues in the various RECQL5 activities. Finally, we have performed a systematic comparison of the RECQL5 structures with other RecQ family structures and based on these comparisons we have constructed a model for the mechano-chemical cycle of the common catalytic core of these helicases.45742314243AbbVieBayer Pharma AGBoehringer IngelheimCanada Foundation for InnovationEshelman Institute for InnovationGenome CanadaInnovative Medicines Initiative (EU/EFPIA) [ULTRA-DD 115766]JanssenMerck Co.Novartis Pharma AGOntario Ministry of Economic Development and InnovationPfizerSao Paulo Research Foundation-FAPESPTakedaWellcome Trust [106169/ZZ14/Z]Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP

The invention features an isolated gene silencing regulatory element that includes 5\u27 TACNNTAC 3\u27. Vectors, transgenic plants and seeds thereof that include such a gene silencing regulatory element are also disclosed. The invention further provides methods of decreasing the transcription of a DNA sequence in a transgenic plant using the isolated gene silencing regulatory element

Economic decision-making in psychopathy: A comparison with ventromedial prefrontal lesion patients

    Get PDF
Psychopathy, which is characterized by a constellation of antisocial behavioral traits, may be subdivided on the basis of etiology: “primary” (low-anxious) psychopathy is viewed as a direct consequence of some core intrinsic deficit, whereas “secondary” (high-anxious) psychopathy is viewed as an indirect consequence of environmental factors or other psychopathology. Theories on the neurobiology of psychopathy have targeted dysfunction within ventromedial prefrontal cortex (vmPFC) as a putative mechanism, yet the relationship between vmPFC function and psychopathy subtype has not been fully explored. In this study, we administered two laboratory decision-making tasks (the Ultimatum Game and the Dictator Game) to a group of prisoners (n=47) to determine whether the different subtypes of psychopathy (primary vs. secondary) are associated with characteristic patterns of economic decision-making, and furthermore, whether either subtype exhibits similar performance to patients with vmPFC lesions. Comparing primary psychopaths (n=6) to secondary psychopaths (n=6) and non-psychopaths (n=22), we found that primary psychopathy was associated with significantly lower acceptance rates of unfair Ultimatum offers and lower offer amounts in the Dictator Game. Moreover, primary psychopaths were quantitatively similar to vmPFC lesion patients in their response patterns. These results support the purported connection between psychopathy and vmPFC dysfunction, bolster the distinction between primary and secondary psychopathy, and demonstrate the utility of laboratory economic decision-making tests in differentiating clinical subgroups
